ABSTRACT

OBJECTIVE@#This study aimed to compare the effects of water storage treatment and thermal cycling on the shear bond strength (SBS) of three self-adhesive dual-cure resin cements.@*METHODS@#Six cubic zirconia specimens with side length of 2 cm were obtained by cutting and sintering. Three self-adhering dual-cure resin cements (i.e., Clearfil SAC, RelyX U200, and Multilink Speed) were selected. According to their bonding modes, they were divided into three groups: direct bonding group (direct coating with resin cement), adhesive group (applying universal adhesives and then coating with resin cement), and primer group (applying Z-Prime Plus and then coating with resin cement). According to experimental conditions, each group was divided into two subgroups: subgroup a (water storage at 37 ℃ for 24 h) and subgroup b (thermalcycling for 5 000 times). SBS data were analyzed by one-way ANOVA by using SPSS 19.0 software (P0.05). After thermalcycling, the SBS of the three types of resin cements decreased (P<0.05); the SBS of the adhesive group was higher than that of the direct adhesive group and the primer group (P<0.05). Fracture mode analysis revealed that the type Ⅲ fracture mode evidently increased after the thermalcycling treatment compared with the water storage treatment.@*CONCLUSIONS@#The universal adhesives and the primer can improve the SBS of self-adhesive dual-cure resin cement in water storage at 37 ℃ for 24 h. The universal adhesives had a better bonding durability than the zirconia primer.

ABSTRACT

To analyze the long-term trend of mortality and years of life lost (YLL) of gastric cancer in Tianjin from 1999 to 2015. From January 1, 1999 to December 31, 2015, the data of gastric cancer deaths was collected from the Tianjin death surveillance system. The inclusion criteria of death due to gastric cancer were coded by using 151 in the 9(th) edition of the international classification of diseases (ICD) (1999-2002) and C16 in the 10(th) edition of ICD (2003-2015). The crude and age-standardized mortality rate (ASR) of gastric cancer was calculated according to Segi's world standard population. YLL was calculated according to the standard method of the disease burden of WHO. The Joinpoint regression was used to calculate the average annual percentage change (AAPC) to indicate the trend of mortality and YLL of gastric cancer. From 1999 to 2015, there were 20 000 deaths of gastric cancer in Tianjin. The proportion of gastric cancer death in the population aged 0-44 years old, 45-64 years old, and 65 years old and above was 4.9%, 30.4%, and 64.8%, respectively. The proportion of males and urban was 67.1% and 67.5%, respectively. From 1999 to 2015, the crude mortality rate was from 12.10/100 000 to 12.58/100 000. The ASR was from 11.04/100 000 to 7.24/100 000. The average annual YLL number was 29 625.83 person-years and the rate was 3.09 person-years per thousand people. From 1999 to 2015, the crude mortality rate and the PYLL of gastric cancer in Tianjin were stable (the AAPC was 0.34% and -0.24%, all values >0.05). The ASR showed a downward trend (AAPC=-2.58%, 0.001). From 1999 to 2015, the ASR of gastric cancer in Tianjin showed a downward trend, and the YLL of gastric cancer was stable.

ABSTRACT

Objective:To construct oxaliplatin (L-OHP) drug-resistant cell line HCT-116/L-OHP in human colon cancer, in order to observe the reversal effect of curcumin (cur) on its drug resistance, and preliminarily explore the possible drug resistance mechanism. Method:The concentration gradient increasing method was used to gradually increase the L-OHP concentration of HCT-116 in parental colon cancer cells, and the cell line HCT-116/L-OHP resistant to L-OHP was established. The cytotoxicity of L-OHP and curcumin to HCT-116 and HCT-116/L-OHP cells was detected by cell counting kit-8(CCK-8) method to observe whether curative resistance could be reversed. Western blot was used to detect the expressions of drug-resistance-related proteins. Real-time PCR was used to detect changes in related genes. Result:Human colon cancer cell line resistant to L-OHP were successfully established and named as HCT-116/L-OHP, with a drug resistance index of 12.6.Compared with HCT-116 cell lines, the expression levels of resected and repaired cross complementation gene 1 (ERCC1) protein and gene in HCT-116/L-OHP cell lines were significantly increased (PP-1), the expression of ERCC1 decreased (PPConclusion:HCT-116/L-OHP cell lines have a stable drug resistance, and its drug resistance mechanism may be up-regulated with the expression of ERCC1, which leads to the up-regulation of Bcl-2,GST-π,MRP,P-gp,Survivin and other related proteins, and enables tumor cells to acquire drug resistance. Curcumin can reverse the drug resistance of HCT-116/L-OHP, and its mechanism may be to reduce the expression of ERCC1, thereby down-regulating the expressions of Bcl-2,GST-π,MRP,P-gp,Survivin and other drug-resistant related genes and proteins, and increase the sensitivity of tumor to L-OHP, so as to reverse the drug resistance of tumor cells.

ABSTRACT

Aim To clarify the role of autophagy in the regulation mechanism of epithelial-mesenchymal transition(EMT) in hepatocytes and the transition to myofibroblast. Methods BNL CL. 2, a mouse embryonic hepatocyte, and BNL CL. 2 cells knocked-down BECN1 genes were treated with TGF-β1(2 μg . L-1) and curcumin(10, 20, 30 jimol • L - 1 ) . Real-time PCR and Western blot were used to detect Beclin-1, LC3, Vimentin and α-SMA gene and protein expression in BNL cl. 2 cells. The optical microscope was used to observe the changes of cell morphology. And the changes in cell autophagy were detected by GFPLC3 plasmid transfection. Results After the intervention of TGF-β1, the expression of Vimentin and α-SMA, the mesenchymal marker, increased and the autophagy decreased. Curcumin could inhibit Vimentin and a-SMA expression in hepatocytes. Moreover, curcumin could increase the autophagy related genes and protein expression of Beclin-1 and LC3 in hepatocytes. Curcumin could increase the level of autophagy and inhibit the cell transformation to myofibroblast-like cells. After interfering with BECN1, the effect of curcumin inhibiting hepatic EMT was partially blocked. Conclusions Curcumin could inhibit the process of EMT of hepatocytes by increasing the level of autophagy. The occurrence and development of liver fibrosis was consequently inhibited.

ABSTRACT

Aim To investigate the effect of broneol on acute lung injury(ALI) induced by lipopolysaccharide (LPS). Methods Male C57 mice were randomly di-vided into saline group, model group, broneol group and dexamethasone group, then the ALI mouse model was induced by instilling intratracheally with LPS. The levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6) and keratinocyte-de-rived cytokine (KC) were measured at 6h, 12h and 24h after instillation of LPS, and the pathological changes of lung were observed. Mice alveolar macro-phages (MHS) and epithelial cells (MLE-12) were stimulated by LPS. After the stimulation of 1h, 3h, 6h,9h, 12h, 24h, the levels of TNF-α and IL-6 in MHS cells and the contents of KC and macrophage in-flammatory protein-2 (MIP-2) in MLE-12 cells were measured. Results Broneol could inhibit the secre-tion of TNF-α,KC and IL-1β;the early effect of bro-neol on IL-6 was not obvious, but the later effect after the treatment of 24 hours was obvious. After LPS instil-lation 6h and 12h,Broneol could significantly improve lung tissue pathological changes. Broneol had no effect on TNF-α secretion of MHS cells, but it obviously af-fected IL-6 secretion in the later stage. In addition, broneol significantly inhibited KC and MIP2 secretion in MLE-12 cells at the later stage of LPS stimulation. Conclusions Broneol can protect LPS-induced acute lung injury. The mechanism may be related to the inhi-bition of the release of inflammatory factors, the activa-tion of inflammatory cells and the aggregation of neutro-phils.

ABSTRACT

cAMP-specific phosphodiesterase type 4 (PDE4) is one of the hot targets for treatment of inflammatory diseases. PDE4 inhibitors can suppress inflammation by increasing the concentration of cAMP in inflammatory cells. The efficacy and safety evaluations of several PDE4 inhibitors are currently carried on in clinical trials, for example GSK256066 in asthma, roflumilast and GSK256066 in chronic obstructive pulmonary disease, tetomilast in inflammatory bowel disease, and apremilast in dermatitis and arthritis etc. This article reviews the recent progress on PDE4-targeted therapy for inflammatory diseases.

ABSTRACT

OBJECTIVETo investigate the effect of air pollution on stroke mortality in Tianjin, China, and to provide basis for stroke control and prevention.

METHODSTotal data of mortality surveillance were collected by Tianjin Centers for Disease Control and Prevention. Meteorological data and atmospheric pollution data were from Tianjin Meteorological Bureau and Tianjin Environmental Monitoring Center, respectively. Generalized additive Poisson regression model was used in time-series analysis on the relationship between air pollution and stroke mortality in Tianjin. Single-pollutant analysis and multi-pollutant analysis were performed after adjustment for confounding factors such as meteorological factors, long-term trend of death, "days of the week" effect and population.

RESULTSThe crude death rates of stroke in Tianjin were from 136.67 in 2001 to 160.01/100000 in 2009, with an escalating trend (P = 0.000), while the standardized mortality ratios of stroke in Tianjin were from 138.36 to 99.14/100000, with a declining trend (P = 0.000). An increase of 10 µg/m³ in daily average concentrations of atmospheric SO₂, NO₂ and PM₁₀ led to 1.0105 (95%CI: 1.0060 ∼ 1.0153), 1.0197 (95%CI: 1.0149 ∼ 1.0246) and 1.0064 (95%CI: 1.0052 ∼ 1.0077), respectively, in relative risks of stroke mortality. SO₂ effect peaked after 1-day exposure, while NO₂ and PM₁₀ effects did within 1 day.

CONCLUSIONAir pollution in Tianjin may increase the risk of stroke mortality in the population and induce acute onset of stroke. It is necessary to carry out air pollution control and allocate health resources rationally to reduce the hazard of stroke mortality.

ABSTRACT

To develop a model for forecasting the mortality of stroke in Tianjin,China. The time series of stroke mortality from 1999 Jan.to 2006 Dec.in Tianjin city were subjected.Circle distribution analysis was used to verify the trend of time concentration.Multiple seasonal autoregressive integrated moving average model [ARIMA (p,d,q) (P,D,Q)s],based on model identification, estimation and verification of parameter, and analysis of the fitting of model, was established. Most of the deaths from stroke occurred in January and had a cycle of 12 months. An AR/MA model (0,1,0)×(0,1,1)12 was established(1-B)(1-B12) lnxt=0.001+(1-0.537 B12)εt.Conclusion: ARIMA & Circle Distribution analysis is an important tool for stroke mortality analysis.Potentially it has a high practical value on the surveillance, forecasting and prevention of stroke mortality.

ABSTRACT

OBJECTIVETo investigate effects of the leaching liquids of 5 different kinds of dental alloys on L929 cells at cell level and molecular level.

METHODSThe fibroblast L929 cells of mouse were cultivated in vitro in leaching liquids of 5 different kinds of dental alloys, Au alloy (n = 8), Ag-Pt alloy (n = 8), Co-Cr alloy (n = 8), Ni-Cr alloy (n = 8), and Cu alloy (n = 8). The RPMI 1640 cell medium containing 10% fetal beef serum was used as control. The cytotoxicities of the 5 dental alloys were evaluated by means of methyl thiazolyl tetrazolium (MTT), and the effects of these alloys on the expression of caspase-3, caspase-8, and caspase-9 mRNA of L929 cells were examined using reverse transcription polymerase chain reaction (RT-PCR) method.

RESULTSAfter 48 hours culture the cytotoxicity of Cu alloy group was in Grade 4 and those of the other groups were all in Grade 0. The mRNA levels of caspase-8 had no change in all groups (P > 0.05). The mRNA levels of caspase-3 were as follows: Cu alloy (0.474 +/- 0.001), the negative control (0.527 +/- 0.003), Au alloy (0.528 +/- 0.013), Co-Cr alloy (0.615 +/- 0.007), Ag-Pd alloy (0.673 +/- 0.009), and Ni-Cr alloy (0.803 +/- 0.037). The mRNA levels of caspase-9 were as follows: Cu alloy (0.532 +/- 0.041), Au alloy (0.574 +/- 0.013), the negative control (0.578 +/- 0.010), Co-Cr alloy (0.617 +/- 0.009), Ag-Pd alloy (0.703 +/- 0.018), and Ni-Cr alloy (0.811 +/- 0.037). There were significant differences between the groups except the negative control group and Au alloy group.

CONCLUSIONSThe Cu alloy shows the highest cytotoxicity, and the leaching liquids of 5 different kinds of dental alloys may induce cell apoptosis through mitochondrion pathway.

ABSTRACT

OBJECTIVETitanium, by far one of the oldest materials in medical implants used for hard tissue replacement and rehabilitation, was used widely as implant material. The objective of the present study was to fabricate the TiN modified layer on the surface of medical pure titanium and improve its surface properties. Thus the wear resistance, biocompatibility and antisepsis were improved.

METHODSA continuous-wave 2 kW Nd: YAG laser was used to irradiate the sample in the environment of N2 with concentration of 99.995%. Titanium nitride surface coatings had been extensively used as corrosion resistant and biocompatible layers on titanium and its alloys. The microstructure and composition of the laser nitrided coating were studied by scanning electron microscopy and X-ray diffraction. Microhardness of the modified sample was also analyzed by micro-hardmeter.

RESULTSTiN modified layer with the thickness of about 400 microm was obtained. TiN distributed gradually from surface to the substrate. There was a good metallurgical bonding between the nitrided layer and the substrate. The hardness of the modified layer was reduced from the surface to the inside.

CONCLUSIONWith optimum process parameters, a compact laser modified gradient coating reinforced with fine TiN was achieved on the surface of medical titanium.
